Solutions – types of solutions (standard, stock, saturated). Expression of concentrations – molarity, molality, normality, mass fraction, mass percent, ppm, ppb – interconversion. Percentage solutions and ratio solutions – problems. Microscopy – Bright field , Dark field , Phase contrast, Fluorescence, Scanning and transmission electron microscopy, confocal microscopy- Flow cytometry. UNIT 2 SEPARATION TECHNIQUES 9 Hrs. Cell disruption methods and Cell fractionation – Homogenization media, High pressure homogenization, Ultrasonication – Protein precipitation, ammonium sulphate fractionation of proteins. Centrifugation: Principle, types and applications- Differential, isopycnic density gradient centrifugation, Rate zonal centrifugation. Filtration: Ultrafiltration, reverse osmosis, membrane filtration, Dialysis. UNIT 3 SPECTROSCOPY AND CHROMATOGRAPHY 9 Hrs. Spectroscopy – Principle, block diagram, interpretation of results and applications - UV-Visible spectrophotometer - Infrared , Fluorescence Spectrophotometry, Atomic Absorption Spectrophotometry - Raman Spectroscopy - Mass Spectrometry, NMR and ESR. Chromatography: Principle, methods, interpretation of results and applications- TLC, Column Chromatography - Ion exchange, Gel Filtration and Affinity Chromatography, HPLC and GC UNIT 4 MOLECULAR TECHNIQUES 9 Hrs. Electrophoresis of nucleic acid and proteins: Principles, methods and applications- Agarose gel electrophoresis, Pulse field gel electrophoresis, Capillary Electrophoresis, SDS – PAGE, IEF, 2D PAGE. Blotting techniques - principle, methods and applications - Southern, Northern, Western, Dot, slot and Zoo blot- Immunological techniques: - Ag-Ab reactions- Immunoelectrophoresis- ELISA, RIA. UNIT 5 TRACER TECHNIQUES AND DIAGNOSTICS 9 Hrs. Tracer techniques- autoradiography – radiodosimetry, XRD. Immobilization of cells and enzymes - supercritical fluids methods- Nano biotechnology in molecular diagnosis: current techniques and applications- Biosensors- principle, methods and applications in biotechnology- microfluidics and Biochips technology.
